Shots were fired into the Yarraville industrial site clubhouse in Campbell St about 12.20am Sunday morning, but the gunmen are yet to be identified.
Victoria Police Det Sen Constable Cameron Pye said more than five shots were fired into the clubhouse although no-one was inside.
But firing shotguns and handguns near residential areas would not be tolerated, he said.
''The potential for people being injured is very high,'' he said.
A warrant was executed to search the clubhouse this morning and members of the Bros were ''reluctant'' to cooperate with police.
CCTV has been seized but is yet to be reviewed.
Police say there is no known motive and were unable to confirm if the shooting was perpetrated by another club or individuals wanting to warn off the Bros.
The Bros in 2011 were caught up in an internal feud between its Yarraville and Melton chapters after the defection of members to a larger outlaw club.
The feud involved their Melton clubhouse being burnt down.
A police operation codenamed Indigo seized drugs and firearms from the Bros clubhouse in May that year.
Raids were conducted on several homes of Bros bikies across Melbourne's west.
The operation focused on the sale and distribution of commercial quantities of amphetamines.
